As a general rule it takes about 200 miles of use for self-adjusting assemblies to get to their optimal braking power. What you can do to speed this process up is take your trailer out to an empty parking lot and apply the manual override on the brake controller several times as you travel at a slow speed.
How do you tell if your trailer brakes are working?
The easiest way to tell if the brakes are working on the trailer would be engage the manual override and have a friend sit outside the vehicle and watch the wheels on the trailer. If they begin to drag when you let off the brake pedal (slowly) on the tow vehicle then you know they are working.

How do self adjusting trailer brakes work?

An electromagnet, connected to an actuating arm, is energized by electric current from the brake controller in the cab of the tow vehicle.

The easiest way is to raise one side of the trailer and spin the wheels while someone applies the surge brakes at the coupler. The better way would be to remove each of the wheels and brake drums and activate the brakes while making sure the wheel cylinders and brake assemblies are functioning properly.
